According to Samuel Luckhurst, the veteran midfielder felt tightness in a muscle issue as he warmed up in the dressing room.
Defender Willy Kambwala will now take a position on the bench. The 19-year-old has played 10 games in all competitions this campaign, including two FA Cup appearances.
Speaking before kick-off, Erik ten Hag said: "Casemiro is not 100 per cent and in that space around Phil Foden we need a strong defence."

Casemiro, meanwhile, has come under intense scrutiny for his performances at centre-back after filling in at the back amid a slate of injuries.
He was slammed following his showing at Crystal Palace as the Red Devils were smashed 4-0 at Selhurst Park.

But the 32-year-old kept a clean sheet against Brighton in his last outing.
Unfortunately for the player, he won't take any part in his side's FA Cup final against Man City.
This season, Casemiro has made 32 appearances across all competitions, scoring five goals.
He's been linked with a move away from Old Trafford this summer, with Saudi Pro League clubs thought to be interested in acquiring his services.
